【Animal Modeling】-Aniline-induced DNA damage in mouse hepatocytes and lymphocytes and its repair effect

  Objective: To study the genotoxicity of aniline and its repair kinetics.

  METHODS: Single-cell gel electrophoresis (SCGE) technique was used to detect the relationship between DNA damage and time-course of hepatocytes and lymphocytes in KM mice after a single gavage of 100 mg/kg aniline for 3, 8, 16, 24, and 32 h.

  Results: The results of SCGE experiment showed that the tail length and tail moment of hepatocytes gradually increased from 8 h, and reached the maximum DNA damage degree at 16 h. Compared with the control group, the difference was significant (P<0.01). 32="" the="" degree="" of="" dna="" damage="" gradually="" at="" h="" p="">0.05); while the tail length and tail moment of lymphocytes gradually increased at 16 h, reached the maximum at 24 h, and the DNA damage gradually recovered at 32 h .

  Conclusion: Aniline has potential genotoxicity to hepatocytes and lymphocytes; the changes of the two DNA damage indexes have a significant time effect relationship, indicating that these two types of cells have an effective DNA repair mechanism.
