[Animal Modeling]-Hematopoietic stem cell transplantation in adult immunodeficient mice

  The hematopoietic stem cells are intravenously injected into the immunodeficient mice established by the radiation image, HSC must remove the T cells to reduce the graft-versus-host immune response. The successful HSC transplantation can rebuild the human immune hematopoietic system in mice. The operation process is as follows:

  Put 5 to 12 weeks old NOG or NSG mice into a sterilized container with a filter. Mice were mapped with 240cGy radiation (137Cs radiation source). Inject human HSC within 4 to 24 hours. Mirroring the mouse can kill mouse HSCs and provide a direction for transplantation of injected human HSCs. Prepare human umbilical cord blood cells, use CD3 antibody magnetic beads to separate and purify CD3+ T cells. The HSC cells in 0.5ml PBS buffer containing 3×10000/CD34+ were injected through the tail vein of the mouse. Human HSCs are maintained in mice for 10-12 weeks, during which time human HSC cells will gradually grow and differentiate to produce human T and B cells. After 4 weeks of HSC injection, human CD45+ cells can be detected, indicating that the human immune system has been successfully rebuilt in mice.
